I am looking at hiding a couple of multi-caches but I haven't done one yet. Are there limits to the number of stages a multi-cache can have? Are there requirements on what actually has to be at each stage? How close can each stage be to each other or far away and do the same 250 ft. rule apply if part of a stage is near an existing cache?

No limits on the number of stages, but remember that if one stage has a problem, you'll need to disable the cache while fixing the problem. The proximity of any physical stage (something you placed) to any other existing physical element of a cache with a different GC# needs to be greater than 528 feet apart (not 250'). Here is a link to the Geocaching Guidelines with some examples.

I have a multicache question... can the first stage of a multi be in a public library where the cacher would need to ask a librarian for a certain book (placed by the CO) in order to figure out the coordinates to the final? The only thing I could find in the guidelines that might put a kibosh on it, under "Must be Assessable" is:
No contact required. Caches cannot require geocachers to contact the cache owner or anyone else.

Caches cannot require geocachers to contact the cache owner or anyone else. Yes, that would be correct. Contact with anyone to locate the cache or information is not allowed. Usually the best way to set up a library cache is to hide a container outside with information about how to locate something inside the library......usually a hollowed out book. I have seen magnetic keyholders also used. Maybe you could just progress this to include one more stage?
